We always rent a car. The quickest way to get there is the B (Bee line) toll road. Costs a buck for toll, takes about an hour and 15 minutes from Orlando to the Space Center. Nice ride, freeway. Check your maps to get on this road. It starts near the airport.

Really this depends on if you want to rent a car or take a tour bus. If you rent a car out to Disney you will take the 417 south loop to hwy 528 (the beachline) then on to KSC. You can also take a tour bus check this website for more info.
